Unfortunately NO, both AMC and Regal utilize Fandango as a merchant/retailer for online purchases. The only place a Regal Gift Card can be used is at the Regal Box Office and counter sells. Fandango has a gift card which can be used online at at Fandango Kiosk.
After you pay for your tickets on fandango, you have to take the credit card that you used to the theater to pick up the tickets. You are also given a confirmation number. You can print that page and redeem it at the theater for the tickets.
