No, you will not die from a lack of sleep. But it is impossible for you to resist your brain's urge to sleep. Your brain releases a chemical called endogenous morphine (one of the primary ingredients of sleeping pills) when it needs to sleep.

It is during sleep that your brain can organize the information retained when you are awake. Your brain also has the opportunity to expedite the regeneration of certain cells which are essential. If you force yourself to stay awake, you will have trouble with memory and focus. You will also start suffering from Psychosis.

The release of endogenous morphine is a safety mechanism to keep you from overloading your brain. You will always fall asleep before real damage can be done to your brain.

And if you were to take drugs like amphetamine to combat your brain's urge to sleep. Your brain would simply release more and more endogenous morphine. And in that case you would die from health issues such as heart attack from the amphetamine or you would fall into a state of unconsciousness (coma) caused by damage to your brain or the excessive amount of endogenous morphine.
