If ONLY the muffler is missing from the rear of the car, and the rest of the exhaust system is intact, then the only damage will be to your hearing, and your wallet in the form of a ticket for defective equipment if you drive it on the street like that. However, I would NOT recommend running it for extended periods like that, as the engine is designed to run with a certain amount of backpressure, and with the muffler gone you are de-tuning your exhaust system and lowering performance. It's not like the old days where running open headers gave your car a power boost. Also you could be getting exhaust in the car itself as the pipe is open before the exhaust exits out from under the car.
