Yes, you can drive over Hoover Dam. In fact, until just recently it was the main highway from Las Vegas to Phoenix, AZ.
In October a new bridge was opened which bypasses the dam. It was needed because the dam had become a major bottleneck.
However, the dam remains a fascinating historic site, and automobile traffic continues for tourism purposes.

There are six of them on the Colorado river. Glenn Canyon dam, Hoover dam, Davis dam, Parker dam, Palo Verde diversion dam and Imperial dam.
It is a 3 hour drive from the Hoover dam to the Grand Canyon viewing areas. The Grand Canyon National Park actually adjoins the Lake Mead Recreational Area which surrounds the reservoir held back by Hoover Dam.
