Once you've beaten her, you can go to the Cave of Being in order to meet with the female Professor Juniper and you'll come across Sinnoh's 3 Pixies (Azelf, Mesprit and Uxie) and then they will depart. Each one will appear somewhere in the region as soon as you step on a specific spot.
For Mesprit, the specific spot you need to step on is on the top of the Celestial Tower, below the stairs.
Additionally if you don't want to catch Mesprit in Black 2 or White 2 then you can use the Poké Transfer Lab in Route 15 in order to transfer a Mesprit you have in a previous game.
