No, she couldn't have as she was the 3rd programmer hired by Howard Aiken to program the Harvard Mark I. The other 2 programmers before her had written many programs already.

This is usually credited to Ada Augusta Byron Countess of Lovelace who wrote a program for Charles Babbage's Analytical Engine, but never ran it as the machine was never built.
