If you're an American, no, you do not, as Puerto Rico is considered a part of the US. However, some airlines and cruises might require you bring one, and the US government recommends (but does not require) that you bring one. The reason being that if for some reason the boat or plane you're on needs to make an emergency stop in a neighboring country like the Dominican Republic or Haiti, you would suddenly need a passport!
