All medical schools in the United States have pre-requisites for admission. Classically these consist of Biology, Chemistry, Organic Chemistry, Physics, English composition or some comparable class, and frequently a Statistics course. Some medical schools are also requiring Anatomy and Physiology classes, Biochemistry, and Microbiology. In addition to these courses, almost all require the Medical College Admissions Test (MCAT), which tests proficiency in many of these subjects.
No, you don't have to be enrolled in a Pre-Med program, but there are definitely a set of "pre-med" courses that are required prior to being accepted to any medical school.

Most four year universities and colleges will have some sort of pre med program. The pre med program is not a major, but advising to help you decide that medical school is the right decision for you and then help you get in and meet the requirements for medical school. Of you want to find a good premed program then look at the percentage of kids that applied to med school and got in.

FAMU doesn't have a medical school but they have the pre-med program.

Although the University of Pennsylvania does not technically have a pre-med program, they have a great reputation for having their students enter med school. Penn has an awesome med-school too & it's an ivy league. This would be my first recommendation. Some other options, Penn State & University of Pittsburgh. Hope this helps!
