Do you need to reach a certain point in heartgold and soulsilver before you can use the pokewalker?
Depends on how you see it, all you need to do is have a Pokemon in a box. So you technically have to have access to pokeballs. Other than that once you have a Pokemon in a box just save and restart the game and the third option at the main menu will be to connect to pokewalker and just follow their instructions.

You can't get a jigglypuff in Diamond. It says on Bulbapedia that you can get a jigglypuff in the trophy garden but it's very rare! Try to trade with someone who has soulsilver or heartgold for a jigglypuff because jigglypuff is there.
