Yes but not as the main character. Normally you play as Yoshi trying to save the rest of your friends (Mario, Luigi, and Wario). Inside the castle you can find paintings with each of the characters pictures on them. Inside each painting is a boss you must defeat in order to save your friend.
After doing so caps will begin appearing on the stages. Grab an M cap to transform to Mario.
Also if you go to where Peach's Secret Slide is you can find an M letter room. If you go inside there you will become Mario.
