No you do not need a battery, but the lights and horn will not work unless you have a battery installed, dead or not.
Just make sure you buy one that, first of all fits the space provided and can be securely mounted. Then make sure it has at least the same CCA as the OEM battery that was installed by Honda. The more CCAs the better especially in cold climates.

You can if the key has a door lock/unlock button. I lock mine all the time with the remote. If you have the button on the key and it is not working then maybe the battery in the remote is dead? Open it up and replace the battery.
