When you click on something you want to attack, then click "attack", the "march" window of your Rally Point opens. In the top right under the word "attack" there is a drop down box for which hero you want to select to lead the attack.
If by "lead" you are talking about appointing as the mayor of your town, this is done by clicking on the Feasting Hall and in the upper left side of the window the "appoint mayor" button is available.
As you play, there are advantages to changing who is mayor, depending on whether you are needing to defend your city in an attack and create troops, or have faster construction times and more production.
