How are the witches of oz related?

No, Glinda isn't the sister of either the subsequent Wicked Witch of the East or of the West. But the Wicked Witches of the East and of the West are half-sisters. Nessarose Thropp is the future Wicked Witch of the East. Elphaba Thropp is the future Wicked Witch of the West. Elphaba and Nessarose share the same mother, but have different fathers.

In the movie the east and west witches were sisters, and Dorothy meets the witch from the south, while the wizard represents the north witch.

In the book Dorothy meets the witch from the north, and the wizard represents a fifth power in the center of Oz.

Yes and no. At first, the Wicked Witch of the West and Glinda don't get along at all in 'Wicked'. They're enemies. But then they realize that others are trying to keep them from being friends. So they're able to work out a mutually acceptable form of friendship.
