In the mission "Doberman" which takes place fairly early on in the game in Los Santos, CJ gets to enter the Amu-nation store for the first time. Sweet calls him to deal with a pusher problem in Glen Park, so CJ is required to go to Amu-nation first to get tooled up, and the gun store is then unlocked.
