There were no series letters on 1990 $100 bills. You may be looking at a plate-position marker or some other security feature. In any case, except for uncirculated bills and those with printing errors, all modern-date $100 bills are worth only face value.
In the 1950s, there were several different series of 10 dollar bills issued by the treasury. The series range from A through E, and have different values depending upon the series. For a series A 10 dollar bill, the value online is between 45 and 100 dollars, depending upon condition.

Besides the consumer methods (watermark, reflective ink, testing the paper), the Treasury can also check the plate numbers against the serial numbers for sequential bills, or do a mass spectrometer test of the ink or paper of the bill. The inks used are specifically designed, are changed slightly with each series, and are nearly impossible for the average counterfeiter to duplicate.
