Yes, you can play Microsoft Flight Simulator without a joystick, you can play with just a mouse or keyboard, but a joystick makes it much easier.

Right click on the screen, at the bottom of the pop up menu there is selection called mouse yoke.

Yes, you can use a joystick with Microsoft Flight Simulator X. I'm not sure of all the kinds but I use a Sidewinder Force Feedback 2 and find it great, it cost about $100.
