It will depend upon the settings of AutoTune. The makers of the software (and originally, hardware) never intended it to be an effect that would be noticed by the listener. Only extreme settings by artists such as Cher in her song 'Believe' and later by artists like T-Pain did the extreme settings (so called 'Hard Tune') become mainstream. The easiest way to tell is to look for notes beyond the range of the singer's voice which are hit exactly, pitch slides that are abnormally abrupt, and the lack of any vibrato on a note.
