At the time of the Ripper murders and for quite some time after, Scotland Yard spoke to and interviewed hundreds of suspects and possible witnesses reguarding the Whitechapel Murders, to no avail. JTR was daring committing his crimes out on the street where he could have been discovered at any moment. But science did not permit his capture and through no fault of the police or the detectives that worked the case the Ripper was never identified.
The ideal situation would be DNA evidence. The problem is there is almost nothing left of the actual evidence. A world war, two relocations, and pilphering of the case records and physical evidence has left very little material to work with. At this point in time, the best way would be to first eliminate as many suspects as possible. That would leave a short list of probable suspects. Without physical evidence though, it would be an educated guess.

Find a good, detailed review of Discovery Channel's "Jack the Ripper in America" at The 44 Diaries blog. It gives you a complete summary of the documentary, and includes many links and photos. Including the new composite of what detectives think Jack the Ripper really looked like.
There is a museum that claims to have the knife of Jack The Ripper. If you 'read the fine print' you will find that it is not the knife used by the Ripper. There was never a weapon recovered in this case. In what remains of autopsy reports of the Ripper's victims it can't even be determined as to the exact type and size of the knife used.
