Isildur lost the One Ring while fleeing from orcs after defeating Sauron. He was ambushed by orcs in the Gladden Fields and was shot by arrows, causing him to drop the Ring as he tried to escape.
It probably would not have affected anything. He was killed because the ring had left him - the ring's fate was more important than Isildur.
