They had thermometers that were a glass tube with Mercury sealed inside. There were temperature markings on the outside of the tube that when the mercury reached that point it showed what the temperature was.

It is not possible to measure the room temperature with a thermometer. A digital oral thermometer will not work to measure the room temperature. It is meant to measure human temperature, not room temperature. You will need a room thermometer to measure the room temperature.

Temperature is the measure of thermal energy. And how you measure thermometers is that if the liquid expands when its temperature increases. The height of a column of the liquid inside the tube shows the temperature.

"What do you measure (when) you measure..." ? When you measure an object's temperature, you are measuring the amount of heat the object emits (gives off). There is no such thing as cold, only the absence of heat.

assuming the temperature is equally distributed you could measure the temperature?

Yes, "thermometers" are instruments used to measure the temperature of things.

Temperature is not a measure of the amount of heat stored in a substance. It is the measure of the kinetic energy of the molecules of a substance.
