I just replaced the left rear door tail light and it was more than I bargained for. I have a 2003 Sienna, but I bet they are similar. I had to first remove the clips that cover the screws on both handholds. Then remove the screws and the handholds. Then, at the base of the door, gently start to pry the door panel off. Set it aside and make sure you have all the clips (some of mine fell out when I removed it and had to be popped back in). Then replace the bulb that is out by rotating it slightly until it drops out and you can pull the bulb loose. I had a little trouble putting the panel on because of the position of the rear defrost wires, but it eventually went on. You just have to press fit it back on all over the door. Then reinstall the handles and cover clips. If you are talking about the side tail lights I know you take out 2 screws and then there are 2 metal clips built into the assembly. It does come off, but you have to find just the right angle to get it.
