You can still mod weapons, but can't create any 'impossible' combinations. Basically you can only mod in a weapon that could actually be found in the game. That still lets you mod a weapon into a one in a million drops 'perfect' form with all the best components, and equal level to yourself. Though not as powerful as pre-patch hack items, it will be considerably stronger than anything you find for many levels after (and you could keep modding it to raise its level too).
No, what they did was patch the game so that 'impossible' weapon combos added by mods, are removed. You can mod in a weapon that 'can' exist as far as the random drops tables are concerned, and that weapon can be duped.

If you're asking if you can change from say the Siren to the Solider in mid-game while keeping your quest progress and levels, no. You'll have to start a new game as one of the other characters. If you're playing Borderlands on the PC, there may be a patch or mod that allows you to do this.
