Well, I haven't tried this yet, but I think you just beat All-star as Zero Suit Samus. You can turn into her before you fight everyone by holding Z before the match starts. It didn't work in classic mode (to turn into her before a match) so it may not work but it's sure worth a shot. I think it only works before a brawl because you're supposed to hold Z at the character screen with your player as samus then hold it until you get to the choose a stage screen and you'll see zero suit samus' head instead of her helmet at the top left corner.

There is no difference, because you do All-Star mode to get the final smash trophies for all of the characters. Completing All star with Every Character is different because Zero Suit Samus is the same character as Samus and Zelda is the Same as Shiek. To collect all final smash Trophies for all characters, you have to play as Samus, Zero Suit Samus, Zelda, Shiek and the other thirty three characters. There is another difference, playing co-op gets final smash trophies but not "completing all-star".
