You can not gain entry until the mission where you have to use Leonardo's flying machine. If you are trying to get into the Assassin Tomb then you should know that its entry is on the church roof next to the Basilicia Di San Marco.

∙ 13y agothe sanctuary is the room where you place all the seals to unlock altair's armour. So to get into the sanctuary you need to go through the bookcase.

∙ 13y agoAfter a certain memory is completed, the bookshelf in Mario's office will open to reveal a hidden room behind it. there is where the sanctuary is located.
