Once you find their HQ, Cabbot will give you a mission to go to the Glow (he will mark it on your map) and retrieve an artifact. You will need anti-radiaton drugs and a rope. The artifact you need is on the first floor, it is a holodisk and its own description will tell you it is the thing you are looking for. However it's worth exploring the old base thoroughly for some nice items and interesting background info.
First you need to find their HQ. One easy way is going to the Hub, you can get their map location from the arms dealer, or from one of the caravans. Once there, speak to Cabbot and ask to join, he will give you a mission to go to the Glow and retrieve an artifact from there. The guard next to him will give you a Rad-X, and it's advisable to have some other anti-radiation drugs with you. Also, take a rope.
When you get to the glow, use the rope to descend into the crater. There are six levels in all with some nice equipment (such as a suit of Combat Armour) but you can get the holodisk you need from the top level. Return this to Cabbot, and you're in.
You go to Windhelm to this little boy named Aventus Aretino in his house you will here gossip around Skyrim about him go to him and do his quest, then find a courier(ex; RiverWood there is one) he will give you a note that has a black hand on it that says "we know" then find a bed go to sleep and you will wake up in anouther room do this part of this quest also and the rest will lead you to the darkbrotherhood

(edit)- im not sure you need the quest but if u walk into riften some guy will give you a quest to talk to Aventus Aretino, when you find the kid he will give you a quest to kill his evil teacher, lol
You need to complete the Quest "The waters of life" then "The American dream" when you report back to Elder Lyons at the Citadel you will be made a Honary Member of the Brotherhood of Steel

Apparently there are two merchants in the game which you can get it from. The 'tough guard' guarding the Brotherhood of Steel initiate in The Hub - The old town.
There is a few things that you will have to do... 1. get done with the quest The Water Of Life 2. beating that quest give you access to the citadel. 3. go into the citadel some where in there you will get a another quest that you must do and after it you can join. (Extra) after you join, you can go into the citadel, there is a person too talk to about, "Power Armour Training." people ask me about how to get it all the time this will let you wear the Brotherhood's armor.
