You can't get through the locked door from the outside, and you can't jump up to the second floor inside. So you have to get the owner to open the outside door.
Doctor Spyglass is actually an inventor working for the spy agency. Let him know that Director D sent you by asking for an eye exam. Deliberately choose the OPPOSITE symbol each time (up-down, left-right, M instead of W, etc.). He will go upstairs and open the outside door.
Go outside and jump to the door. Dr. Spyglass will give you the Chameleon Suit.

When he points to a symbol, select the one that faces the opposite direction (backward E instead of E, M instead of W). Failing it like that shows that you are a friendly spy. He will go upstairs, and now you can exit to find that the upstairs door is unlocked.
