I don't know what "2rs" means, but, 600 10th Avenue is between West 43rd and 44th Streets. Penn Station is at 33rd to 34th Streets, between 8th and 7th Avenues.

You can walk the whole way: north to 43rd Street, then west to 10th Avenue: 7th Avenue ---> 8th Avenue ---> 9th Avenue ---> 10th Avenue.

Or, you can take the Uptown A-C-E (the blue line), 1 stop, from 34th Street-Penn Station (at 8th Avenue) to 42nd Street-Port Authority Bus Terminal (at 8th Avenue). Then walk 1 block north to 43rd Street, and 2 blocks west to 10th Avenue.
