First you have to be "friends" with them on Facebook. Then, go to your profile and click on the "Info" tab. To the right of the "About me" section, there should be a "edit" button to click on. Once you do that, there's a list on the left side, one of which should say "relationships". In there, you can "add family members" by searching their name and entering their relation to you. A request will be sent to them and will become official once they approve.
Additionally, you can change the privacy setting so that some people see the relationships and others do not. If you go back to "Info" and "edit", on the right below the list there should be the words "privacy settings" in blue. Click on that and you can choose who is able to see those relationships or not.
