Only supervisors can remove flags from questions (or answers). If you feel a question or answer has been flagged wrongly - contact the category supervisor (they're show on the right of the screen) - via their message board, with a request to have the flag removed. The supervisor will usually look at the question - and remove the flag if necessary.

Indeed you can. If you want to edit or remove flags, go to your Site tools, and there should be something marked "Flagged questions". Click that, and you will be directed to all the flag questions, where you can remove, edit, or add flags onto questions and/or answers.

No, flags that you removed before joining The Initiates Program do not count towards your total for the removing flags mission. Similarly, any flags that you removed whilst on a previous mission do not count towards your total either. It is only the flags that you remove from the start date of your mission that count towards the mission. You can check how many flags you have removed by clicking on "My contributions" on the blue menu and filtering to "Flags".
