Yes, they will run simultaneously, but why? Defraggler will reorganize and defrag your files, while CCleaner gets rid of some. So run CCleaner first then after the garbage has been taken out, run Defraggler to clean up your house. There is no point in defragging garbage.

I assume you mean Comodo System Cleaner. it is not as comprehensive as cCleaner and does not have a uninstall utility or a way to block startup programs as cCleaner does. I run cCleaner and highly recommend it. It is very safe to use and is the best free one you can get.
CCleaner only works with the Windows operating system and will not work with Mac OS X. As the Mac OS is a completely different system most of what CCleaner does would not apply to the Mac OS. If you are running the Windows operating system on a Mac then CCleaner could be used.
