Usually the tail will tell you. Also the spots on the bottom of their shell, but the easiest way to tell is look at its tail. If it looks rather small and thin, it's a girl. If it's fat and long, it's a boy. -Chris
Female red eared sliders have a smaller tail but are physically larger. Males have longer front claws. The cloaca of a male is closer to the end of the tail.
if your turtle is a male you will see a bump on either side of its tail. if its a female its more likely to be aggressive and will only have a slight bump on either side of it's tail.
