The best thing to do to unlock characters is to beat each and every cup, in all difficulties (50, 100, 150 & mirror) in first place.
I think there's at least one or two characters that need to be unlocked by beating the normal staff ghosts in the time trials, too.
Also, to unlock Mii Suit B, I think you need to either beat every expert staff ghost, and/or get a triple star rating in each cup on all difficulties.
