How does one get wrinkles out of a thick vinyl projection screen?

Use a hair dryer! Put the hair dryer on high and aim it at the wrinkles. Let the natural weight of the screen pull the softened vinyl into place. Let the straightened screen cool completely before re rolling. Do not fold the screen again, roll it up.

You can use a blow dryer.
It will warm the fabric and that will get wrinkles out.
The level of success will depend on how bad the projection screen is wrinkled.
When using a blow dryer, make sure not to put it too hot, as it will melt the fabric. Keep a safe distance as well, don't hold it against the fabric.
When done gently and with care, it gets great results. Impatience will lead to damage to the screen, as is using it a full power.
