It depends on the route you take. Once you're across the bridge in downtown Niagara falls NY, follow the signs to go over Grand Island. This is the quickest route--about 25 minutes if the bridges' traffic is moving quickly. There's a toll for the bridge, though. Alternatively, you can drive down highway 62, which becomes Niagara Falls Blvd when you enter Erie County. If the traffic is okay, it will take about 35-40 minutes to get to the edge of the actual city of Buffalo. It depends partly on where in Buffalo you want to go.
