Rhode Island was unique in that it offered religious freedom. Most other colonies had a state-sponsored or "Established" church. Within that colony, you had to practice the "established" religion, or you would be barred from certain activities such as voting or running for political office. In Maryland, for example, the Roman Catholic religion was the "Established Church" and non-catholics were discriminated against.

Rhode Island did not have an established church, and did not discriminate against anyone. For example, the first Jewish Synagogues in the New World were in Newport, Rhode Island.

On a side note, you may recall the curious phrasing of the First Amendment to the Constitution of the United States. It was worded that way because no colony wanted to have the Federal government make any changes to their "Establishments of Religion". So the Bill of Rights prevents the Federal government from establishing its own church, but does not prevent the individual States from having their own established churches.
