Concrete never fully cures, but a non-quicksetting concrete should be firm on the surface in about 4-8 hours, and usable for foot traffic or post setting in 36 hours.
Pre-mixed concrete is beter as it made to BSI standards and hand mixed (dry bagged) can be weakend by an inexperianced person mixing it. But it depends on what you are using the concret for; if its just a slab for a garden shed to go on it dose not realy make much differance, but if its structural you best get it pre-mixed. I do believe it would depend on the relative humidity and ambient temperature as concrete takes over 100 years to cure properly
