I took too many Dramamine by accident once, and it was not pleasant. I don't recommend it as something to do for "fun", it wasn't.

Ask the pharmacist questions about all drugs and remedies you buy at the drugstore, part of their jobs is to provide consultation to patients. Even if prescribed, don't take medicines until you have researched them thoroughly and read the package information. Relying on other people for information like this (e.g., asking for anonymous people online to answer) instead of relying on doctors or pharmacists for information is a crapshoot, and testing drugs on yourself is stupid.
Okay. Here goes. If you are talking about diphenhydramine, and you are an adult the recommended dosage is usually around 25-50 mg every 6-8 hours. Just read the label on the bottle. If you are talking about dimenhydrinate, and you are an adult then take 1-2 tablets every 4 to 6 hours; not to exceed 8 tablets in 24 hours. Children 6-12 years of age, should take 1/2- 1 tablet every 6-8 hours; not to exceed 3 tablets in 24 hours. Children 2-6 years of age, take 1/4 -1/2 tablet every 6-8 hours; not to exceed 1 1/2 tablets in 24 hours. Now by tablets, I mean to say 50mg of Dimenhydrinate. This is the usual dosage of one tablet of Dimenhydrinate. Believe me when I say this, do not take more Dramamine then you need to, it is not a good idea and it can have some nasty side effects.

I'm pretty sure you take Dramamine so that you do not get nauseous. You can take it before long plane and car rides to keep from getting sick too.
I travel every week. On Saturday morning, I take two dramamine less drowsy (that's a total of 50mg meclinzine). I'm not fully normal until the following Wed.

In my personal experience with Dramamine it takes an hour for me to feel its total effects. However if you don't take Dramamine that often, then if you take it a half hour before departure you should feel it while departing. I'd like to also add that taking dramamine 24 hours before your departure will do nothing. The effects will have been long gone. It wont be more effective just take it a half hour to an hour before your departure.
I'm going to say no. I get very car sick and took Dramamine for a long time. I started taking Zoloft about two years ago. this past spring break i used the patch version of Dramamine and it made me absolutely worse. I was sweating, shaking, virtually in tears. I called my dr and he told me to take the patch off. When i took it off i could feel the difference immediately. i wasn't shaking, sweating or anything. I still had to sit in the front because of my car sickness, but NO don't take Dramamine if you are taking Zoloft
