I have checked 5 websites and come up with a different number on each one. I went to Google.es, which is the google version for Spain and looked for a definition for ciudad (city) in Spanish and couldn't find one.
I did find the related link which lists municipalities by population. There is one list of 397 cities by name with a population of 20,000 or more.
The third table from the bottom is titled
Distribución de la población por tamaño de municipiosThis table is a breakdown of all the municipalities in Spain by population range beginning with cities of more than 1,000,000 inhabitants and ending with municipalities with a population of less than 100 inhabitants.The best I can suggest that you pick a minimum number of inhabitants that you consider necessary for a municipality to be a city as opposed to town, village etc. and add up the number of municipalities the meet or exceed your definition.
