The earliest settlers of the Plymouth Colony set sail on the Speedwell ship from Delfshaven, Holland in July 1620. They sailed to Southampton, England to meet with the Mayflower ship and the two left for the new world on August 15, 1620. They had to turn back when the Speedwell began taking on water. Some of the passengers were chosen to travel on the Mayflower from Plymouth, England and they finally left on September 16, 1620. The Mayflower reached land on November 21, 1620.

The Mayflower and the Speedwell were the ships the first Pilgrims sailed on. Both set sail for the New World on August 15, 1620, but were forced back twice due to dangerous leaks on the Speedwell. The Speedwell's passengers regrouped on the Mayflower, and on September 16th, the voyage began, a month behind schedule. The Mayflower landed near what is now Plymouth, Massachusetts in 1620.

The Pilgrims set sail from England on two ships. Which ship did not make it to the New World?
Actually, they set sail on only one ship, the Mayflower. You may be thinking of the Speedwell which brought some Pilgrims to Southampton harbor to board the Mayflower. You may also be thinking of the other ships that settled the Virginia colonies (before the Mayflower set sail and landed in Plymouth). The Susan Constant, the Godspeed, and the Discovery all set sail from England together and arrived in what is now Virginia in 1607. The Mayflower (1620), the Fortune (1621) the Anne (1623) and the Little James (1623) all went to Plymouth and those on board were the early Plymouth Colony settlers.
