Technically we don't NEED different rooms in a house... for most of human existence, the majority of people have gotten by with single-room dwellings ... but given the option, most people prefer to have at the bare minimum a bathroom which is distinct from the room(s) in which they cook, eat, and/or sleep.
Castles had many many rooms such as: A chapel, hall, watch tower(s), Kitchen(s), bedrooms (at first chambers), Toiletrooms, bathrooms and a keep they're the ones i know of if i find anymore ill improve it :)

You can only increase the size of your rooms (and add more rooms) by speaking to Tom Nook and paying off subsequent debts. There's a limit to how many rooms you may have in your house as well as the largest available size before you're considered debt-free though - for example, you're only allowed six rooms total: front room, back room, right wing, left wing, basement and attic. The largest size is the same as your front room after the initial expanding process whereby Nook allows you to specify which room(s) to add or improve.
