Absolutely. Driving nonstop exept for bathroom, dinner stops and fuel it would take you 3 days to drive it one way. Take I-10 east to I-15 north to I-40 east to I-44 east to I-70 east to the Pennsylvania Turnpike to NJ Turnpike north to Holland Tunnel. With that much time left over you can make lots of interesting side trips and take a different route on the return journey.

The total cost is around $25 if you drive into Manhattan. The first three tolls are 2, 3 and $4 each, the NJ Turnpike depends on where you get off.($7-$8 approx.), and the Lincoln and the Holland Tunnel entering NYC (Manhattan) are both $8. You only have to pay to enter NYC and not to return. You will only enter one Tunnel and it depends on which part of NYC you are heading to whether you take the Lincoln or the Holland Tunnel.
As with most other toll roads, the charge on the Massachusetts Turnpike depends on how far you drive on the road.
