Yes, series the two batteries to produce 12 volts. Tap off of the two end terminals for the 12 volts. Tap off of the series jumper for the 6 volts of one of the batteries. The two 6 volt batteries can not be paralleled in this connection as they will short out.
300 watts is not any volts. Watts are how much power is being used regardless of the voltage. For example you could have a 300 watt spot light in a truck running off the 12 volt system in the truck or you could have a 300 watt security light on your house running off of 120 volts.

Watts = Amps x Volts. Amps = Watts/Volts. 10000/120 = 83.3 amps can be used at 120 volts. On a generator that large it is most likely that it also has the capacity to produce 240 volts. 10000/240 = 41.6 amps can be used at 240 volts. What you can connect to the generator will be totals of the amperage of devices that do not go above these amp ratings.
It's the amps that are controlled by the breaker not the volts. You can have a 600 volt 15 amp breaker, you can have a 347 volt 15 amp breaker. The breaker will trip when you exceed 15 AMPS.
