Once the mother weans her current litter, she'll be ready to mate again.
It takes an average of 68 days or between 56-74 days for them to give birth. Larger litters are due in smaller days and small litters are due in longer days. So the to 'how often a guinea pig can breed' is kind of up in the air. I'd say probably 3-4 litters per year.

Guinea pig pregnancies last 59 - 72 days. Litter size can be from 1 to 10 approximately. The female (sow) is fertile immediately after giving birth to the babies, and if the male is present (boar), she could very possibly get pregnant again. NOTE: This is NOT recommended, as it could put a large strain on the female (feeding the current litter at the same time as pregnant with the new one is not considered a delightful situation for the sow).

You should wait ATLEAST 4 1/2 months. Advised is 6 months. Back to back breeding for a sow is very dangerous, even deathly. They need to restore their health and weight to the normal, before mating and having another litter. If breeding like this is not done, your sow will most likely pass along to the Rainbow Bridge.

Right after the babies have weaned off the mom. However the mom will get stressed if you do this and could die, It is recommended to wait a month or two (the longer the better).
