Widal test is done by qualitative method or quantitative method.
Quantitative method:-Take a set of 8dry clean test tubes.
Place 1.9ml saline in tube no.1 and 1ml saline in rest of tubes. Add 0.1ml serum in 1st tube mix well it is dilution of 1:20
Transfer 1ml diluted serum from tube no.1 to tube no. 2 and so on.
Through tube no.7 discard 1ml of fluid after mixing.
leave 1ml saline in tube no.8.
Add one drop of appropriate antigen(use only that antigen suspension which has given a positive reaction in screening test.
Mix well and incubate overnight at 37.c
Examine for agglutination.
Interpretation:- Only a titre above 1:80 should be considered as significant
A rising titre (done each week)is considered to be definite evidence of infection.
positive or negative with approximately titre.
normal value of widal test Only a titre above 1:80 should be considered as significant. A rising titre (done each week)is considered to be definite evidence of infection.
Dreyers tube is narrowtube with a conical bottom for H agglutination which helps to see loose cottony wooly clumps at bottom. Felix tube is round bottom used for O agglutination and positive result appears as disc like pattern at the bottom of the tube.

What is false positive in widal test
widal slide is easier and it save time but widal tube test is not easy and it takes time.
I mean widal test result positive after treatment
Isolated Widal test reading does not have much clinical significance. Rising titre of Widal test done one week apart, in a febrile patient, indicates typhoid fever. You can have such rising titre of Widal test in healthy patient, after dose of typhoid vaccine.
1:80 titre is significant for determination of widal positive.
WIDAL
widal test
To a certain extent yes.The post antibiotics Widal Test will have Low titers of agglutinins.Hence the test may be reported as False Negative.
widal test.
manual test of widal method
In Widal test, you measure the antibodies. It takes minimum one week to develop the antibodies of typhoid bacteria.
Widal test is negative in incubatory carrier. Widal test is always positive in chronic carrier.
