Only use 1X PBS. 10X PBS is made up because it is easier to store and can be diluted when needed. Labs that use PBS often use it in large quantities so making up 10 or 20 liters at a time is usual and making it up as a concentrate is easier to do.

Molarity of 1X PBS is 10mM. To prepare 20mM PBS take 16g of NaCl, 0.4 g KCl, 2.88 g Na2HPO4 0.48 g KH2PO4 in 800 ml of distilled H2O. Adjust the pH to 7.4 with HCl. Add H2O to 1 liter.
