If you open the 4 back rooms (using an escape rope, flash, water stone, and Ho-Oh), catch all 26 unown, and talk to the researcher in the building it will unlock the ! and ? unown. Catch them and talk to the guy again. This will complete the unown side quest, but that's about it. Nothing really special unless you just really have to have all 28 unown.

Answer Altough in Diamond and Pearl, the unown you catch when shown to the boy that you find in the ruins then walks off to his house near by he will give you ball seals (stickes according to the unown you show him, which you can stick on pokeballs) also once you've caught a-z a cave on the route to the left off the city is acessable and then your able to catch ! and ?
