Depending on if you are using one or two handed weapons, it would probably be one of the following:
- Rune of the Nerubian Carapace (Only for 1h weapons) -- Increases your defense by 13% and your total stamina by 1%.
- Rune of the Stoneskin Gargoyle (Only for 2h weapons) -- Increases your defense by 26% and your total stamina by 2%.
Of course, if you are already at defense cap, then you might want to consider:
- Rune of Swordbreaking (Only for 1h weapons) -- Increases parry chance by 2% and reduces disarm duration by 50%.
- Rune of Swordshattering (Only for 2h weapons) -- Increases parry chance by 4% and reduces disarm duration by 50%.
- Rune of Spellbreaking (Only for 1h weapons) -- 2% spell deflection and reduces silence duration by 50%
- Rune of Spellshattering (Only for 2h weapons) -- 4% spell deflection and reduces silence duration by 50%
If you are dual wielding, you might consider a combination of Swordbreaking and spell breaking. This will give you an additional 2% parry chance, 2% spell deflection, silence duration and disarm duration reduced by 50%.

In most cases, high-end tanks nowadays are Frost/Unholy, but once the next expansion comes, Blood will be designated permanently as the tanking tree for Death Knights.
