The difference lies in the fact that the rough ER has ribosomes lining it and the smooth does not. This also changes the functions of the two types. The smooth ER has diverse metabolic functions, such as synthesizing lipids, detoxifying alcohols and drugs, as well as metabolizing carbohydrates. The rough ER creates secretory proteins, which are usually glycoproteins.
