It depends on the circumstances. If you know you are going to use it multiple times in the day, then you should just leave it on (but in sleep mode) and turn it off at night. If you plan to only use it every few days, then you should turn it off.

With the newer computers the "Sleep" (stand by) mode works fine. If it's "days" before you use it again then i usually just turn it off. The computer runs a "Maintenance" program around 2:00 ish in the morning so it doesn't hurt to leave it on overnight once in awhile if you don't use it very often. It won't do the maintenance if the computer is asleep tho. So disable the "Automatically Sleep" option for those days. You can still use the "Display Sleep" no problem. I believe it will still do the maintenance if you turn the "Put hard disk to sleep when possible" mode in the Preferences/Energy Saver. It just pauses the hard drive so it's not spinning until you need it.
well is it goes to sleep you should just be able to shake the mouse and wake it up but when you get it on look throw your setting to make sure that is not why it is doing it and if that is not it then i don't know what to do because if you can get it out of sleep mode then you cant do much
