Not really, but it depends on how you define "two-party system". The parties in the US have varied over time. The current party system in the US dates from about the Civil War era. That is fairly old by political party system standards, but the UK division between Liberals and Conservatives (or Whigs and Tories) dates back to earlier.

The US does have an unusually stable and consistent two-party system, but given the changes have occured over the years (eg. the Republicans were once the party of the North, and the Democrats of the south, while now it is closer to the other way around), it's not really accurate to say that it is the oldest anywhere.
